Abstract N-9-(2-amino)ethyl-O6-benzylguanine(4) was obtained through substitution and Gabriel reaction, using O6-benzylguanine as the starting material. N-(2chloroethyl)-N′-2-(O6-benzyl-9-guanine)ethyl-N-nitrosourea(6) was synthesized by isocyanate substitution of 4, followed by a nitrosation reaction. 4 and 6 were new compounds. And their structures were characterized by UV- Vis, 1H NMR, 13C NMR, IR and HR-ESI-MS.
